Why CO2 is required to culture cells?

we use to incubate cell culture at 37*c in CO2 incubator.

1. depends on what you are trying to culture. You probably are working with either an autotroph that requires CO2 to create organic molecules, or you are working with an anaerobic bacteria and the CO2 displaces the O2 so they can live/thrive.
2. Co2 is used to gas the culture medium to the correct pH.
